Intelligent siRNA, Dr. Tobias Pöhlmann, Gründerzentrum Jena
„Intelligent siRNA’s” are modified siRNA molecules that could be used in vitro as well as in vivo to modulate gene expression selectively and exclusively in target cells. Due to binding of specific peptides, siRNA molecules become inactivated, and are handled in the inactive form. The short peptides are bound to the ends of the siRNA backbone, which prevents the initiation of the RISC.

The peptides bound on the siRNA consist of specific target sequences for peptide cleaving enzymes, which are expressed specifically in target cells. In these cells, specific enzymes cut off the peptides from the inactivated siRNA, which converts it to the activated form. In non-target cells, specific enzymes are not expressed, so that the „Intelligent siRNA” remains inactive and does not modulate gene expression.

Due to the siRNA structure, up to four different peptides can be bound to the siRNA, which allows an increase of selectivity of the mechanism. It means that after in vivo injection, in some non-target cells, one or two peptides may be cut off, but the „Intelligent siRNA” remains still inactive as long as one single peptide is still bound. Only target cells are able to cut off all peptides and activate siRNA mechanism.

The mechanism of „Intelligent siRNA” can be combined with established methods of siRNA cell delivery such as TAT-Protein binding or receptor/ligand systems. In fact, by binding these components on the inhibiting peptide, these systems are cleaved from the siRNA with the activation process and thereby they have a less influence on the induction of RNAi.
